1090 Extracellular vesicles from IFN-gamma-primed induced mesenchymal stem cells improve atopic dermatitis in mice

Atopic dermatitis (AD) is a chronic inflammatory skin disorder characterized by immune dysregulation, pruritus, and abnormal epidermal barrier function. Compared with conventional mesenchymal stem cell (MSC), induced pluripotent (iPSC)-derived (iMSC) recognized as unique source for producing extracellular vesicles (EVs) because it can be obtained in scalable manner an enhanced homogeneity. In this study, we investigated whether, EVs from IFN-γ stimulated of iMSCs have immune-regulatory, anti-inflammatory, tissue-repairing potential mice model AD. After DNCB was applied to the dorsal NC/Nga three weeks, AD symptoms were assessed. Then treated IFN-γ-iMSC-EVs or JAK inhibitor (Baricitinib) four weeks. AD-like demonstrated application mice, such increased score, scratching number, serum IgE level, thickness, infiltration. Also, decreased expression IL-4Rα, IL-13Rα1, TSLP, their corresponding intracellular signaling molecules activated mice. Furthermore, atopic which supported reduced infiltration mast cells . Impaired barrier, evidenced upregulation keratin, filaggrin, ceramide synthase, also observed that received IFN-γ-iMSC-EVs. The results our study may contribute developing novel cell-free therapeutic strategies currently undergoing phase 1 clinical trials patients dermatitis.
